Transcription factors regulating blood oxygen linked to melanoma metastases

Apr. 16, 2013 ? Researchers at the University of North Carolina have discovered that transcription factors regulating the levels of oxygen in the blood also play a role in the spread of the skin cancer melanoma.

In research published April 8 in the Journal of Clinical Investigation, a research team led by William Kim, MD, member of the UNC Lineberger Comprehensive Cancer Center, and graduate student and first author Sara Hanna, linked melanoma metastases to a pair of transcription factors known as HIF1 and HIF2.

Researchers found that HIF1 and HIF2 are overexpressed in melanoma tumors. In healthy cells, HIF1 and HIF2 assist in regulating hypoxia, the state caused by low levels of oxygen in the blood. Hypoxia has been linked to metastases in several sold tumors, and the UNC team has found that it promotes the spread of melanoma from the skin to other sites in the body through the lymphatic system.

Patients who are diagnosed with early stage melanomas have a high rate of survival, but the prognosis worsens significantly once the tumors spread to other sites throughout the body. Using in vitro systems and mouse models, researchers suppressed the expression of HIF1 and HIF2 in the melanoma tumors. While the inactivation of the transcription factors did not reduce the growth of the initial tumors, it did reduce the rate at which the melanoma spread to other sites in the body.

Both HIF1 and HIF2 independently activate the protein kinase SRC using different signaling pathways. The SRC protein has been linked to several different cancers, and the identification of its role in melanoma suggests that existing therapies targeting SRC may prove to be a viable target for therapies aimed at reducing the spread and ultimate lethality of the cancer.

"What we are trying to do now is inhibit these pathways with drugs in the mice to see if we see a decrease of metastasis," said Hanna.

Monarch butterflies are well known for their ability to fly 2,000 miles south from North America to Mexico each fall and back again in the spring. Now, researchers reporting in Current Biology, a Cell Press publication, on February 21 have evidence to show that the butterflies would just keep on heading south if it weren't for the chilly weather.

The findings help to explain why the butterflies spend the winter on frosty mountaintops. They also imply that global climate change could profoundly influence the monarchs' migrations, the researchers say.

"The monarchs need the thermal microenvironment at the overwintering sites for the migration cycle to persist," said Steven Reppert of the University of Massachusetts Medical School. "Without that thermal stimulus, the annual migration cycle would be broken and we will have lost one of the most intriguing biological phenomena in the natural world."

Earlier work by Reppert's team found that monarchs rely on an internal compass and skylight cues to guide them in their long-distance travels south. Patrick Guerra and Reppert have now found that the butterflies rely on the same system for their return flight north.

They next wondered what triggers the switch in the butterflies' overall direction. To find out, they captured fall migrants at the start of their migration, subjecting them in the laboratory to the same changes in temperature and light that they would experience in the mountains of Mexico. Remarkably, after 24 days of such treatment, the butterflies headed north upon release.

Further study confirmed that the switch in flight direction depended only on the experience of cold, not on changes in day length. They also confirmed that butterflies brought in and protected from the cold kept right on flying south.

Given these discoveries and the recent availability of genetic and genomic tools for monarchs, Reppert said his team is now poised to understand much more about the biological processes underlying the iconic migration. And that could make all the difference for the monarchs' future.

"This increased understanding will help us protect the migration," Reppert said.

Hey, don't mean to interrupt what you guys got going here just wanted to say I look forward to reading what you guys come up with :] Worlds Finest couldn't be a better title for Superman and Batman! I just finished reading the Justice League Reboot, which was awesome btw. It's nice to see some DC love instead of the massive Avengers/Marvel boners everyone seems to be sporting right now lol Ok, done buggin you two, write on! ;D

The Music Tapes Announce Circus Tent Tour - "The Traveling Imaginary"

In support of their new album, Mary's Voice, The Music Tapes (Julian Koster, Robbie Cucchiaro, The 7-Foot-Tall Metronome, a legion of singing saws, and friends) are embarking on a unique US tour. Called The Traveling Imaginary, the band will be heading to towns and cities across the country and pitching the circus tent home that they built specially for this tour for evenings of songs, stories, games, movies, magic, amusements, and more.

The tent, which was funded by The Music Tapes? recently successful Kickstarter campaign, has been a longtime dream of Julian?s. On entering the completed structure for the very first time, he said, ?If you made a circus tent out of blankets in your room when you were a little kid and then fell asleep, it's like the dream you would have.?

The Music Tapes have a long history of hosting surreal tours since they first emerged as part of the influential Elephant 6 Collective in the late 1990s ? including the annual Caroling and Lullaby tours that have brought them into over 500 homes across the country. The Traveling Imaginary is an extension of those shows on a more ambitious scale than anything The Music Tapes has ever done.
